¥includes advice for (1) construct design, (2) pronuclear injections (2 times), (3) provision of tail tips for genotyping. Note that on average 3 transgenic lines can be produced from two injections. * Knockout mice: includes (1) advice for construct design, and screening strategy, (2) electroporation of the targeting construct (up to 3 times), (3) selection of ES clones for screening, (4) Cre and/ or Flp electroporation of the targeted ES cells, (5) karyotyping of the ES cell clones , (6) expansion and freezing of clones; (7) blastocyst injections (4 times) for 2 targeted ES cell clones; (8) provision of C57 Bl/6, 129 Svev mice for germline transmission tests; (9) provision of ROSA26 mice for testing cre-transgenic mice; (10) provision of Flipper mice for Flp recombination The PI will be responsible for making the targeting vectors, genotyping/Southern blot analysis of the picked ES clones, and genotyping mice using tail tips provided by the TCF. Cost will be less if only a subset of the above is required, please contact Professor Cheah for discussion. The cost is to cover consumable supplies, maintenance of TCF infrastructure (e.g. breeding banks, foster mothers, germline mutant production, husbandry etc), animals and part of the staff salary cost. Booking method: Each PI wishing to embark on a knockout/transgenic project should first consult Professor Cheah who will advise on design of project, vector design and grant application if applicable. She and the TCF will provide the track record for the project proposal. PIs should put the cost into the budget of their research proposal and ensure they have sufficient funds to cover the cost. |
